HinT hydrolyzes lysyl-AMP, an activity that may be involved in the regulation of translation or the scavanging of aminoacyl-adenylates (Chou, 2006). hinT is essential for growth under high salt conditions (Chou, 2005). Hydrolyzes purine nucleotide phosphoramidates, including adenosine 5'monophosphoramidate (AMP-NH2), adenosine 5'monophosphomorpholidate (AMP-morpholidate), guanosine 5'monophosphomorpholidate (GMP-morpholidate) and tryptamine 5'guanosine monophosphate (TpGd) (PubMed:15703176, PubMed:20934431). Hydrolyzes lysyl-AMP (AMP-N-epsilon-(N-alpha-acetyl lysine methyl ester)) generated by lysine--tRNA ligase, and lysyl-GMP (GMP-N-epsilon-(N-alpha-acetyl lysine methyl ester)) (PubMed:15703176). Is essential for the activity of the enzyme D-alanine dehydrogenase (DadA) and is required for E.coli to grow on D-alanine as a sole carbon source (PubMed:21754980). Is also required for growth at high salt concentrations (PubMed:15703176).
