Applicable Applications for IL1RL1 cell lysate
Western Blot (WB)
Application Notes
WB: Use at an assay dependent dilution.
Species
Mouse
Recommend Usage
1. Centrifuge the tube for a few seconds and ensure the pellet at the bottom of the tube.2. Re-dissolve the pellet using 200uL pure water and boil for 2-5 min.
Sequence Construction
A DNA sequence encoding the mouse ST2 (NP_001020773.1) (Met1-Arg332) was expressed with a C-terminal polyhistidine tag.
Preparation Method
Cell lysate was prepared by homogenization of the over-expressed cells in ice-cold modified RIPA Lysis Buffer with cocktail of protease inhibitors. Cell debris was removed by centrifugation. Protein concentration was determined by Bradford assay (protein assay, Microplate Standard assay). The cell lysate was boiled for 5 min in 1 x SDS loading buffer (50mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 5% b-mercaptoethanol, and lyophilized.
Preparation and Storage
Store at 4 degree C for up to twelve months from date of receipt. After re-dissolution, aliquot and store at -80 degree C for up to twelve months. Avoid repeated freeze-thaw cycles.
Samples are stable for up to twelve months from date of receipt.
Related Product Information for IL1RL1 cell lysate
IL-1 receptor-like 1 (IL1RL1) is a membrane receptor involved in TH2 inflammatory responses and eosinophilia. It has previously been described that levels of the interleukin-1 like 1 (IL1RL1) protein can be used to identifies cardiovascular disease and determine the prognosis for a patient with cardiovascular disease. The ligand for IL1RL1 has been described and named IL-33. Mutants in IL1RL1 have been associated with blood eosinophil counts in a genome-wide association study and with asthma in family-based and case-control studies. As an important mediator involved in many immune and inflammatory responses, this cytokine has been implicated as a regulator of both the development and effector phases of type 2 helper T cell responses, and as a negative feedback modulator of macrophage proinflammatory function. IL33 is a specific ligand of ST2L and induces the production of Th2 cytokines.

This Mouse ST2/IL-1 RL1 overexpression lysate was created in HEK293 Cells and intented for use as a Western blot (WB) positive control. Purification of ST2/IL-1 RL1 protein from the overexpression lysate was verified.

Uniprot Description

IL1RL1: receptor for IL33. Its stimulation recruits MYD88, IRAK1, IRAK4, and TRAF6, followed by phosphorylation of ERK1 and/or ERK2, p38-alpha, and JNK1. Apparently promotes the activation and differentiation of T-helper 2 cells. Belongs to the interleukin-1 receptor family. Highly expressed in kidney, lung, placenta, stomach, skeletal muscle, colon and small intestine. Three isoforms of the human protein are produced by alternative splicing. Isoform A is prevalently expressed in the lung, testis, placenta, stomach and colon. Isoform B is more abundant in the brain, kidney and the liver. Isoform C is not detected in brain, heart, liver, kidney and skeletal muscle.

Protein type: Membrane protein, integral; Receptor, cytokine

Cellular Component: proteinaceous extracellular matrix; extracellular space; cell surface; membrane; integral to membrane; plasma membrane; extracellular region; external side of plasma membrane

Molecular Function: interleukin-1 receptor activity

Biological Process: negative regulation of cell proliferation; negative regulation of T-helper 1 type immune response; negative regulation of interferon-gamma production; negative regulation of I-kappaB kinase/NF-kappaB cascade; signal transduction; positive regulation of interleukin-5 production; positive regulation of macrophage activation; positive regulation of inflammatory response

Product Notes

The IL1RL1 il1rl1 (Catalog #AAA8117085) is a Cell Lysate produced from HEK293 Cells and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 317aa. AAA Biotech's ST2/IL-1 RL1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). WB: Use at an assay dependent dilution. Researchers should empirically determine the suitability of the IL1RL1 il1rl1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: Met1-Arg33 2. It is sometimes possible for the material contained within the vial of "ST2/IL-1 RL1, Cell Lysate" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
