Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate GP130 in samples. An antibody specific for GP130 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyGP130 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GP130 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GP130 bound in the initial step. The color development is stopped and the intensity of the color is measured.

gp130: gp130 is a ubiquitously expressed type I cytokine family receptor. The receptor systems for IL6, LIF, OSM, CNTF, IL11 AND CT1 utilize gp130 for initiating signal transmission. Binds to IL6/IL6R (alpha chain) complex, resulting in the formation of high-affinity IL6 binding sites, and transduces the signal. Does not directly bind IL6. May have a role in embryonic development. Contains 5 fibronectin type III domains and 1 immunoglobulin-like C2-type domain. Two alternatively spliced isoforms have been described.
