These translocations were associated with aberrant transcripts in which 4 predicted zinc finger domains, contributed by the widely expressed chromosome 13 gene ZNF198, were fused to the FGFR1 tyrosine-kinase domain. Transient expression studies showed that the ZNF198-FGFR1 fusion transcript directed the synthesis of an approximately 87-kD polypeptide, localizing predominantly to the cytoplasm.
The novel sequence that was fused to FGFR1, comprising the 5-prime end of the fusion transcript, was mapped to chromosome 13 with the CEPH mega-YAC library and the Genebridge 4 radiation hybrid panel. The findings were consistent with the pericentromeric 13q11-q12 assignments for the t(8;13) cytogenetic breakpoints.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ZMYM2 in samples. An antibody specific for ZMYM2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyZMYM2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZMYM2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZMYM2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a protein that contains nine MYM-type zinc finger motifs. Expression of this gene may mediate the inhibition of hematopoietic cell development during ontogeny, and the encoded protein may also play a role in transforming growth factor-beta signaling as a Smad binding protein. The translocation between FGFR and ZNF198 (8;13)(p11;q12) is oncogenic, causing stem cell leukemia lymphoma syndrome (SCLL), a myeloproliferative disorder (MPD) that evolves to acute myeloid leukemia and/or lymphoma. The transforming activity of ZNF198-FGFR1 requires the proline-rich domains, but not the ZNF domains, of ZNF198.
